In view of the increasing of Hp antibiotic-resistance, the duration and kinds of antibiotics applied in clinical, this study focused on the clinically effective drug Jinghua Weikang Capsule and studied the bacteriostatic and bactericidal effects against antibiotic-resistant Hp in addition with the mechanism of this drug and its major ingredient Chenopodium Ambrosioides. The study found in vitro Jinghua Weikang Capsule and its major ingredient Chenopodium Ambrosioides had bacteriostatic and bactericidal effects against Hp standard strain NCTC1637&26695 and antibiotic-resistant strains with dose-effect relationship, and these effects were synergy with the metronidazole and clarithromycin. Chenopodium Ambrosioides could reduce the multiple antibiotic resistance of Hp, of which the the molecular biological mechanisms might be decreasing the efflux pump genes expressions. In vivo, after repeated exploration and attempt, the animal models of mice infected by Hp SS1 strain and metronidazole-resistance strain 26695 16R were successfully established respectively, through the pretreatiment of immunosuppression creatively. The bactericidal effect which was significantly synergy with the triple therapy including PPI , metronidazole & clarithromycin was found in the Hp SS1 strain infected mice. Due to the experiment is still ongoing, the relative results in the Hp 26695 strain infected mice remains to be observed. The above results confirm the bacteriostatic and bactericidal effects against Hp standard and antibiotic-resistant strains of Jinghua Weikang Capsule and Chenopodium Ambrosioides, and revealed the mechanisms of Chenopodium Ambrosioides against multiple antibiotic-resistant Hp might be decreasing the efflux pump genes expressions. Based on these results, 1 article was published on SCI magazine, along with 4 articles were published or accepted by domestic core journals. Two PhDs and two postgraduates were trained during this research. This study provides the evidences for integration of traditional Chinese and Western medicine treating antibiotic-resistant Hp infection, and the basis for further researches on traditional Chinese medicine against Hp, which is of great significants.
